Detection Of Disease Components Using Magnetic Particles And Microfluidics

US Patent:
20210109094, Apr 15, 2021
Filed:
Oct 12, 2020
Appl. No.:
17/068152
Inventors:
- Cleveland OH, US
Brian Grimberg - Cleveland OH, US
Umut Gurkan - Cleveland OH, US
Susann Brady-Kalnay - Cleveland OH, US
Robert Brown - Cleveland OH, US
Yuncheng Man - Cleveland OH, US
Ran An - Cleveland OH, US
International Classification:
G01N 33/543
B01L 3/00
G01N 21/64
Abstract:
Disease components can be detected in a fluid using magnetic particles and microfluidics. Magnetic particles are combined with a sample in a fluid. The sample may include disease components. The magnetic particles can be configured to tag any disease components within the sample. The fluid can be forced into a microchamber with two microcompartments. In the first microcompartment, the fluid can be exposed to a magnetic field and/or magnetic field gradient. The tagged disease component can be trapped by the magnetic field and/or magnetic field gradient due to the magnetic particles, allowing nonmagnetic components of the fluid to be washed away while the tagged disease components remain trapped by the magnetic field and/or magnetic field gradient. Then, the disease components can be forced into a more narrow second microcompartment and detected using optical instruments.

Epithelial Cancer Evaluation Using Beta Defensin

US Patent:
20210088521, Mar 25, 2021
Filed:
Oct 16, 2020
Appl. No.:
17/072076
Inventors:
- Cleveland OH, US
Santosh Ghosh - Mayfield Heights OH, US
Umut A. Gurkan - Cleveland OH, US
International Classification:
G01N 33/574
G01N 33/543
C12Q 1/68
C12Q 1/6886
B01L 3/00
Abstract:
A method of detecting epithelial cancer is described that includes the steps of: (a) determining the level of beta defensin 3 (BD-3) and beta defensin 2 (BD-2) in a suspect sample obtained from a subject; (b) comparing the level of BD-3 to BD-2 determined in the suspect sample to obtain a suspect BD-3/BD-2 ratio, (c) comparing the suspect BD-3/BD-2 ratio to a healthy BD-3/BD-2 ratio to obtain a diagnostic BD-3/BD-2 ratio; and (d) characterizing the subject as having epithelial cancer if the diagnostic BD-3/BD-2 ratio is greater than 1. A microfluidic device for detecting epithelial cancer using the diagnostic BD-3/BD-2 ratio is also described.

Dielectric Sensing For Sample Characterization

US Patent:
20200405173, Dec 31, 2020
Filed:
May 8, 2020
Appl. No.:
16/870565
Inventors:
- Cleveland OH, US
Pedram Mohseni - Cleveland OH, US
Debnath Maji - Cleveland OH, US
Evi Stavrou - Cleveland OH, US
Umut Gurkan - Cleveland OH, US
International Classification:
A61B 5/05
A61B 5/145
G01N 27/02
G01N 27/08
G01N 27/22
G01N 33/49
Abstract:
As one example, a fluid monitoring apparatus includes a dielectric microsensor that includes a capacitive sensing structure integrated into a microfluidic channel. The microfluidic channel includes a fluid input to receive a sample volume of a sample under test (SUT). A transmitter provides an input radio frequency (RF) signal to an RF input of the microsensor. A receiver receives an output RF signal from the microsensor. A computing device computes dielectric permittivity values of the SUT that vary over a time interval based on the output RF signal. The computing device may determine at least one permittivity parameter based on the computed dielectric permittivity values over at least a portion of the time interval.

Inventors:
- Portland OR, US
Umut Atakan Gurkan - SHAKER HEIGHTS OH, US
Muhammad Noman Hasan - CLEVELAND HEIGHTS OH, US
James Daren Bledsoe - Portland OR, US
Tyler Witte - Portland OR, US
International Classification:
G01N 27/447
G01N 21/64
G01N 33/68
Abstract:
A diagnostic system that includes a cartridge and a reader is discussed herein. The cartridge can contain a patient sample, such as a blood sample. The cartridge is inserted into the reader and the patient sample is analyzed. The reader contains various analysis systems, such as an electrophoresis detection system that uses electrophoresis testing to identify and quantify various components of the blood sample. The reader can process data from the various patient sample analysis to provide interpretative results indicative of a disorder, condition, disease, or infection of the patient.
